Description
EndoTrap® HD is intended for in vitro quantitative removal of lipopolysaccharide (LPS) from biological samples in aqueous solutions such as proteins, antibodies, cell extracts and nucleic acids. EndoTrap® HD can be used in small scale processes for R&D and large scale processes, like manufacturing. It can be applied in early or late biomanufacturing process steps. EndoTrap® HD is based on hydrophilic, dimensionally stable affinity matrix with excellent pressure/flow characteristics.
Affinity chromatography for highly efficient endotoxin removal

Choosing between EndoTrap Red and HD
Buffer Compatibility
Endotoxin removal with EndoTrap® HD works effectively in the pH range of 4-9 and NaCl concentrations of 50-600 mM. Buffers such as HEPES, TRIS, MOPS, MES, and PIPES are recommended. Citrate buffers and chelators of divalent cations (like EDTA) have to be avoided
Note: Customer specific buffers must contain 0.1 - 1 mM Ca2+ or Mg2+. When using PBS buffer always add 1 mM Ca2+ (e.g. CaCl2 ) and 1 mM Citrate pH 7 to the customer specific buffer. Otherwise phosphate and Ca2+ form an insoluble complex and will precipitate.
Leakage ELISA
EndoTrap® affinity chromatography is one principal method for cleaning biological solutions from endotoxin contaminations. The EndoTrap® ligand is a protein by nature, which is bound to the polymeric bead-matrix by stable covalent bonds. However, leakage of minute amounts of ligand is a matter of fact for all affinity materials and testing on these contaminants is often required for regulatory purposes. Depending on the intended use of the preparation and the step in the purification process (early or late), where EndoTrap® is used, a quantitative analysis of residual EndoTrap® ligand might be required. The EndoTrap® HD Leakage ELISA was developed to allow an accurate and reproducible determination of small amounts of EndoTrap® ligand in biological samples. This ELISA is suitable for detection of leached ligand from EndoTrap® HD resin.
